In the present study, we investigated difference in salt stress responses of two barley cultivars (Sunwoo and Sangnok). From six days after seeding, barley plants were stressed with NaCl whose concentration was gradually increased from 50mM to 400mM. Shoot dry weight and SPAD value decreased gradually as soil salinity increased. The value of those parameters was more reduced by salt stress in cv. Sunwoo compared to cv. Sangnok. In order to obtain detailed insight into the protein components associated with the different salt stress response of the cultivars, we have embarked on a gel-based proteomics approach with the 3rd leaf as source material. For this, we have optimized an immobilized pH gradient (IPG)-based two dimensional (2D) gel electrophoresis-based separation of the extracted proteins solubilized in a modified lysis buffer. Preliminary experiment showed a dramatic increase in the solubility of barley leaf proteins and increased spot resolution on 2D gels. Further proteomics analysis on the salt stress-regulated variation of protein spots and their identification will be discussed.
