Revealing transcribed genes of bentgrass species (Agrostis spp.) using sequence related amplified polymorphism.

<>Sequence related amplified polymorphism (SRAP) is informative, abundant and useful molecular genetic marker system used in plants. The objective of this work is to study presence and nature of SRAPs in cDNA of three bentgrass species (Creeping bentgrass, velvet bentgrass, and colonial bentgrass). Fifty-six SRAP primers were used to amplify and sequence discrete fragment bands of cDNA. A total of forty-nine unique fragment bands were isolated from these species. Different SRAP primer combinations used in this study produced different fragment bands. Sequence analysis of cDNA clones and analysis of putative translation products revealed that most encoded amino acid sequences were highly diverse across the grass species used in this study. Sequence analysis of cDNA clones indicated the usefulness of SRAPs for comparative genome analysis of transcribed genes in the grass species. These cDNA sequences are readily available for use in genomic-assisted selection for improved resistance to biotic and abiotic stresses in grass species.
